A hybrid model derived by combing the differential Mueller matrix formalism and Mueller matrix decomposition method for an anisotropic sample containing the linear birefringence (LB), linear dichroism (LD), circular birefringence (CB), circular dichroism (CD) and depolarization properties (Dep) has been developed in our group. This study applies the hybrid model to an optical coherence tomography (OCT) for getting in-depth cross-sectional images of a complex sample with LB, LD, CD and Dep parameters. Thus, the LB parameter related to stress information can be totally decoupled, and the stress analysis in a complex sample like biotissue is possible. In contrast, a simple sample like a polymer with only LB parameter can be tested by the conventional polarization-sensitive OCT.Moreover, the hybrid model can be employed into the optical diagnosis system to investigate the pairs of cancer cell lines. The multiple optical parameters of metastatic cancer cells are evaluated. It is found that highly metastatic cancer cells have the higher LB and Dep properties because of the higher material concentrations inside and more spindle shapes. Thus, it provides a promising method with potential to quantitatively classify the pairs of low-metastatic/high-metastatic for early diagnosis in vitro or ex vivo.
